I see my folders on the TV, but if I move some new folders or files on my PC into this folder, I can't see them on TV anymore. I can only see the old ones. I also did the refresh, still no results.

Make sure that when you add the folders, the TV application is not running. Close everything down and then add the folders, perform the refresh and you will then be able to browse through them. On a side note, make sure that the folders don't have illegal characters such as ASCII codes or non-supported characters.

Samsung does not have its PC Studio application available for Linux operating system. WINE is said to work with Windows applications but in this case, there is the case of the drivers and WINE can’t handle drivers that well in a Linux environment.
I have troubles in trying to connect my TV to SAMSUNG PC Share Manager. My TV is not shown in the "Share" window in Share Manager. Is it possible to set up the DLAN connection if my TV is connected to the router through a cable, and the PC is connected to the router using a wireless connection?

Yes, it is possible. If your TV is not appearing in the "Share" window, I suggest you remove SAMSUNG PC Share Manager from your computer (Control Panel > Uninstall a program) and install it again. After that, use the user guide provided by the developers to set up the connection. If you still have issues, use the contact information from the official web page to get in touch with the support team.
If you use Windows XP or bellow you can go to Start > Control Panel > Add or Remove Programs. In the Currently installed programs box, click on the program and then click Remove. If you have Windows Vista or above you need to go to Start > Control Panel > Programs > Programs and Features. Select the application and click on Uninstall.
If this doesn't work try using Revo Uninstaller. Even if it can't uninstall the application normally, it will force it to uninstall by wiping out all traces of its presence from the system.
After that, try running AdwCleaner to make sure that there are no leftover rogue registry keys or toolbars in your system.

From what it seems, SAMSUNG PC Share Manager is not available for public downloading. Also it's not listed on the downloads page of the official web page. The only way to acquire the software is to contact the developer. After you receive the program, you can install and configure it using the instructions provided by Samsung.

Samsung offers a guide for the DLNA (the process of sharing content with other devices). I recommend you download this PDF guide to your computer. After this procedure, open the file and read about the correct procedure of sharing folders and how to make them available on the Blu-ray device.
